ABSTRACT

Objective: Individuals with schizophrenia and substance use disorders have a poor prognosis and increased psychiatric symptoms. The present study aimed to explore the association of 106 genes in individuals with schizophrenia and comorbid substance use through a next-generation sequencing (NGS) analysis and different in silico algorithms. Methods: We included 105 individuals diagnosed with schizophrenia and a family history of schizophrenia, of whom 49 (46.67%) presented comorbid substance use. Using NGS, we sequenced 106 genes previously associated with schizophrenia. Logistic regression models were used to assess differences in allele frequencies, and a generalized gene-set analysis was performed at the gene level. Functional annotations were performed using different algorithms and databases. Results: We identified a total of 3,109 variants, of which 25 were associated with schizophrenia and comorbid substance use and were located in regulatory and coding regions. We found low-frequency variants in COMT p.Ala72Ser, independently of p.Val158Met, that were associated with substance use. The endocannabinoid functional variant FAAH p.Pro129Thr was also associated with substance use. Conclusions: Genetic variants of genes related to dopaminergic and cannabinoid neurotransmitter systems were associated with comorbid substance use in schizophrenia. Nevertheless, more studies with larger sample sizes are needed to confirm our findings.

ABSTRACT

Angelicae Sinensis Radix excels in activating blood, but the scientific mechanism has not been systematically analyzed, thus limiting the development of the medicinal. This study employed the computer-aided drug design methods, such as structural similarity-based target reverse prediction, complex network analysis, molecular docking, binding free energy calculation, cluster analysis, and ADMET(absorption, distribution, metabolism, excretion, toxicity) calculation, and enzyme activity assay in vitro, to explore the components and mechanism of Angelicae Sinensis Radix in activating blood. Target reverse prediction and complex network analysis yielded 40 potential anticoagulant targets of the medicinal. Gene Ontology(GO) term enrichment and Kyoto Encyclopedia of Genes and Genomes(KEGG) pathway enrichment analysis indicated that the targets mainly acted on the complement and coagulation cascade signaling pathway to exert the anticoagulant function. Among them, the key enzymes thrombin(THR) and coagulation factor Xa(FXa) in coagulation cascade and thrombosis were the drug targets for thromboembolic diseases. At the same time, molecular docking and cluster analysis showed that the medicinal had high selectivity for FXa. According to binding free energy score, 8 potential active components were selected for enzyme activity assay in vitro. The results demonstrated that 8 components inhibited THR and FXa, and the inhibition was stronger on FXa than on THR. The pharmacophore model of 8 active compounds was constructed, which suggested that the components had the common pharmacophore AAHH. The ADMET calculation result indicated that they had good pharmacokinetic properties and were safe. Based on target reverse prediction, complex network analysis, molecular docking and binding free energy calculation, anticoagulant activity in vitro, spatial binding conformation of molecules and targets, pharmacophore model construction, and ADMET calculation, this study preliminarily clarified the material basis and molecular mechanism of Angelicae Sinensis Radix in activating blood from the perspective of big data, and calculated the pharmacology and toxicology parameters of the active components. Our study, for the first time, revealed that the medicinal had obvious selectivity and pertinence for different coagulation proteins, reflecting the unique effect of different Chinese medicinals and the biological basis. Therefore, this study can provide clues for precision application of Angelicae Sinensis Radix and the development of the blood-activating components with modern technology.

ABSTRACT

Objective:To analyze the binding ability of motifs in the serine/threonine kinase StkP extracellular region (EC-StkP) of Streptococcus pneumoniae to β-lactam antibiotics. Methods:Three motifs (SXXK) in the EC-StkP were mutated into AXXA, respectively or simultaneously. Four mutant plasmids (EC- stkp-AXXA1, EC- stkp-AXXA2, EC- stkp-AXXA3 and EC- stkp-AXXA4) were transfected into recipient cells for cloning and expression. SDS-PAGE combined with gel image analysis was used to detect the expression of the recombinant mutant proteins (EC-rStkP-AXXA1, EC-rStkP-AXXA2, EC-rStkP-AXXA3 and EC-rStkP-AXXA4). The expressed mutated proteins were extracted and purified by Ni-NTA affinity chromatography. The binding abilities of the mutant proteins to penicillin (PCN) and cefotaxime (CTX) were detected by isothermal titration calorimetry (ITC 200) and surface plasmon resonance (Biacore t200). Results:PCN and CTX could not bind to the expressed proteins with mutations in the first or the third motif (EC-rStkP-AXXA1, EC-rStkP-AXXA3, EC-rStkP-AXXA4). EC-rStkP-AXXA2 could weakly bind to CTX, but not to PCN.Conclusions:All three motifs in the EC-StkP of Streptococcus pneumoniae could bind to β-lactam antibiotics with the first and the third motifs being more important.

ABSTRACT

The outcome of total hip arthroplasty (THA) in dogs is directly related to surgical planning. Templating of radiographs prior to THA should help the surgeon anticipate prosthesis size and femoral shape allowing canal fill of the proximal metaphysis by the implant ensuring primary stable fixation. The canal flare index (CFI) obtained from radiograph has been used as a measure of risk of complications for the technique in human beings and dogs. However, standard radiographs only provide limited data for the selection of cementless prostheses and the assessment of their fit within the femoral canal, due to factors like radiographic magnification and femoral rotation. Therefore, three-dimensional evaluation based on computed tomography (CT) may be a better tool for CFI measurement. The aim of this study was to compare anatomical measurement with CFI values obtained from craniocaudal radiography and CT. Craniocaudal radiographs using a horizontal radiographic beam (CR), CT, and anatomical macroscopic measurements (A) were obtained from 45 femurs from 23 canine cadavers. The differences between the values of CFI obtained from radiograph (CFI-R), computed tomography on transverse (CFI- TT) and longitudinal axis (CFI-TL) compared to the CFI obtained from macroscopic measurements - gold standard - (CFI-A), and 95% limits of agreement (LOA) between the values, were evaluated by the Bland-Altman method. Dimensions obtained from CT techniques had a greatest mean difference from anatomical and CFI values were also different (P=0.032). Under the experimental conditions, the craniocaudal radiograph, provided the most accurate measurement of the CFI (mean difference: 0.087 ± 0.42).(AU)


O resultado da artroplastia total do quadril (ATQ) em cães está diretamente relacionado ao planejamento cirúrgico. O templating radiográfico pré-operatório da ATQ deve ajudar o cirurgião a prever o tamanho da prótese e o formato do fêmur, o que permitirá um preenchimento ideal da metáfise proximal pelo implante, garantindo, assim, fixação primária estável. O índice de alargamento do canal (Canal Flare Index - CFI) obtido em radiografias tem sido utilizado como fator de risco de complicações para a técnica em humanos e cães. No entanto, as radiografias podem fornecer apenas dados limitados para a seleção de próteses não cimentadas e a avaliação do seu encaixe no canal femoral, devido a fatores como ampliação radiográfica e rotação femoral. Portanto, a avaliação tridimensional baseada na tomografia computadorizada (TC) pode ser uma ferramenta vantajosa para a mensuração do CFI. O objetivo deste estudo foi comparar a medida anatômica com os valores de CFI obtidos na radiografia craniocaudal e na TC. Radiografias craniocaudais utilizando feixe radiográfico horizontal (CR), tomografia computadorizada e medidas macroscópicas anatômicas (A) foram obtidas de 45 fêmures de 23 cadáveres caninos. As diferenças entre os valores de CFI obtidos na radiografia (CFI-R), na tomografia computadorizada no eixo transversal (CFI-TT) e no eixo longitudinal (CFI-TL), em comparação com os valores de CFI obtidos nas medições macroscópicas - padrão-ouro - (CFI-A) e os limites de concordância de 95% (LOA) entre os valores, foram avaliadas pelo método de Bland-Altman. As dimensões obtidas pelas técnicas de TC apresentaram maior diferença média dos valores anatômicos, e as do CFI também foram diferentes (P=0,032). Nas condições experimentais, a radiografia craniocaudal forneceu a medida mais precisa do CFI (diferença média: 0,087 ± 0,42) para representar o padrão-ouro deste estudo.(AU)

ABSTRACT

OBJECTIVE:To study the e ffects of stilbene glycoside c(TSG)on phosphorylation of Thr 205,Ser404 sites of Tau protein in Aizheimer ’s disease (AD)model mice ,and to investigate the potential anti-AD mechanism of TSG. METHODS :APP/ PS1/Tau three transgenes (3×Tg-AD)mice were randomly divided into model group ,positive control group (huperzine,0.15 mg/kg),TSG low-dose ,medium-dose and high-dose groups (0.033,0.1,0.3 g/kg),with 6 mice in each group. In addition ,6 C57BL/6J mice were chosen as normal control group. Administration groups were given relevant medicine intragastrically. Model group and normal control group were given equal volume of normal saline intragastrically ,once a day ,for consecutive 60 days. After last medication ,immunofluorescence staining was used to detect Tau protein and phosphorylated Tau protein (Thr205, Ser404 sites) distribution and expression in brain tissue of mice in each group. Western blotting assay was used to detect phosphorylated Tau protein (Thr205,Ser404 sites)expression level in brain tissue of mice in each group. RESULTS :Compared with normal control group ,the expression of Tau protein,phosphorylated Tau protein (Thr205,Ser404 sites)in 729011126@qq.com the brain tissue of mice were increased in model group ,which were easy to aggregate and distributed more widely ;theirrelative expression were increased significantly (P<0.01). Results of Western blotting assay showed that the expression levels of phosphorylat ed Tau protein (Thr205,Ser404 sites)were increased significantly (P<0.01). Compared with model group ,the expression of Tau protein ,phosphorylated Tau protein (Thr205,Ser404 sites) in the brain tissue of mice were decreased in positive control group and TSG groups ;aggregation decreased,distribution narrowed and their relative expression were decreased significantly (P<0.01). Results of Western blotting assay showed that the expression levels of phosphorylated Tau protein (Thr205,Ser404 sites)were decreased significantly (P< 0.01). Compared with positive control group ,There was no significant difference in the distribution of Tau protein ,phosphorylated Tau protein (Thr205,Ser404 sites)in the brain tissue of mice in TSG groups ;the relative expression were not statistically significant(P>0.05);but Western blotting assay showed the expression levels of phosphorylated Tau protein (Thr205 site)in TSG medium-dose and high-dose groups as well as the expression levels of phosphorylated Tau protein (Ser404 site)in TSG groups were decreased significantly (P<0.05 or P<0.01). CONCLUSIONS :TSG can play an anti-AD effect on AD model mice by down-regulating the expression of phosphorylated Tau protein (Thr205,Ser404 sites)in brain tissue.

Background: Total hip replacement surgery is one of the most advanced and technically demanding surgeries in orthopaedics in which blood loss is inevitable. Most of the patients are elderly with multiple co morbidities and the estimated one-year mortality is about 25%. Objective of the study was to assess the need of post-operative blood transfusion in total hip replacement surgery in patients given tranexamic acid versus control. Methods: It was a randomized controlled trial. Sixty patients of both sexes undergoing total hip replacement surgery included in study on the basis of inclusion and exclusion. Equal patients 30-30 (50%) divided in two groups Tranexamic Acid group and Control Group based on a computer-generated random number table. The Tranexamic Acid group received two doses of 10mg/kg body weight of tranexamic acid just before surgery and two hours later intravenously, whereas Control group received two doses of 10mg/kg body weight of normal saline at similar intervals. Numbers of blood transfusions required postoperatively were noted based on the postoperative haemoglobin readings. Results: Mean post-op haemoglobin for the Tranexamic acid group was 10.1±2.6 g/dl and for the Control group it was 9.1±2.4 g/dl. Seven patients (23%) in intervention group required blood transfusion compared to 13 (43%) in control group. Conclusions: Our study revealed that Perioperative Tranexamic acid is a safe and useful method of reducing intra op bleeding and need of post-operative blood transfusion postoperatively in THR patients.

Objective To investigate the sub-cellular distribution correlation between activated LIMK1 (pLIMK1Thr508) and Aurora-A in mouse oocyte meiosis,and changes in Aurora-A location and spindle structure in condition of LIMK1 inhibition.Methods Immunofluorescence staining was employed to detect the sub-cellular localization of pLIMK1Thr508and its spatial-temporal correlation with spindle organizing regulator Aurora-A in mouse oocyte meiosis; BMS-3, the specific inhibitor to LIMK1 activity, was applied to analyze the effects of LIMK1 inhibition on Aurora-A distribution and spindle formation. Results At meiotic prophase,pLIMK1Thr508was weakly detected and concentrated in the germinal vesicle(GV) in oocytes,with no signal of Aurora-A across the cytoplasm and nuclear area;as meiotic assumption approaching,pLIMK1Thr508left nuclear,aggregating as a single dense dote in the vicinity of nuclear, and being co-localized with the emerging Aurora-A; After germinal vesicle broke down (GVBD), pLIMK1Thr508and Aurora-A remained overlapped and concentrated as multi foci around the condensed chromosomes;at metaphase Ⅰ(MⅠ) and metaphase Ⅱ(MⅡ), pLIMK1Thr508was co-localized with Aurora-A on spindle poles;During anaphase Ⅰ(AⅠ) to telophase Ⅰ(Tel Ⅰ) progression, pLIMK1Thr508was detached from spindle poles and mainly concentrated on the cleavage furrow,while Aurora-A loosely congressed on spindle. In ad-dition, LIMK1 inhibition with BMS- 3 destroyed Aurora-A polar location and spindle formation. Conclusions pLIMK1Thr508is a microtubule organizing center (MTOC)-associated protein, may participate in spindle assembly and maintenance through regulating Aurora-A in mouse oocytes during meiotic progression.

ABSTRACT

Objective To investigate the correlation between Streptococcus pneumoniae (S.pneumoniae) StkP kinase and drug resistance and to analyze the binding ability of StkP extracellular region (EC-StkP) to β-lactam antibiotics.Methods A stkP gene knockout (ΔstkP) mutant was constructed from S.pneumoniae strain ATCC6306 by insertional inactivation method.E-test was performed to detect the minimum inhibitory concentrations (MIC) of penicillin (PCN) and cefotaxime (CTX) against ΔstkP mutant and its wild-type strain.Bioinformatic softwares were used to predict the EC-StkP of S.pneumonia strain ATCC6306,to generate the three-dimensional structure model of EC-StkP and to analyze the correlation between the structure and functions of EC-StkP.PCR was performed to amplify the extracellular segment of stkP (EC-stkP) gene and the product of it was sequenced after T-A cloning.A prokaryotic expression system of EC-stkP gene was constructed.SDS-PAGE in combination with a gel image analysis system was used to detect the expression of the recombinant EC-StkP (EC-rStkP).The expressed EC-rStkP was extracted by Ni-NTA affinity chromatography.The binding abilities of EC-rStkP to PCN and CTX were detected by isothermal titration calorimetry (VT-ITC) and surface plasmon resonance (Biacore).Results S.pneumonia strain ATCC6306 was sensitive to PCN (MIC=0.06 μg/ml) and CTX (MIC=0.12 μg/ml),but its ΔstkP mutant was resistant to the two antibiotics (PCN MIC=16 μg/ml,CTX MIC=32 μg/ml).The 295 aa segment was predicted as the extracellular region at C-end of StkP of S.pneumoniae strain ATCC6306,containing four penicillin-binding proteins and Ser/Thr kinase-associated (PASTA) domains.The cloned EC-stkP segment and the EC-stkP segment in GenBank shared 99.6% similarity in nucleotide sequence and 100% in amino acid sequence.The constructed prokaryotic expression system for EC-stkP gene expressed EC-rStkP in soluble form.Both PCN and CTX could bind to EC-rStkP and CTX was better than PCN in term of binding ability.Conclusion The stkP gene of S.pneumonia is closely related to drug resistance and the encoded protein,Ser/Thr kinase StkP,can recognize and bind to β-lactam antibiotics.

Objective@#To assess the association of the FSHR Thr307Ala-Asn680Ser gene polymorphism with male infertility.@*METHODS@#We searched Pubmed, EMBASE, Web of Science, CNKI, and WANFANG databases for literature on the correlation of the FSHR Thr307Ala-Asn680Ser gene polymorphism with male infertility published from 2005 to the present time. According to the inclusion criteria, we included 12 epidemiological case-control studies and subjected them to a comprehensive analysis with the Stata11.0 software.@*RESULTS@#A total of 2 893 male infertility patients and 3 312 controls were involved in the 12 studies. The Thr307Ala (rs6165) gene polymorphism was shown to be a risk factor for male infertility among the three comparison models (homozygous comparison model, hybrid comparison model and dominant comparison model), with the pooled odds ratios (OR) of 1.26 (95% CI: 1.03-1.54, P = 0.023), 1.18 (95% CI: 1.03-1.36, P = 0.018), and 1.20 (95% CI: 1.05-1.37, P = 0.006), respectively. And the Asn680Ser(rs6166) polymorphism was a risk factor for male infertility in the homozygous comparison and recessive comparison models, with the pooled ORs of 1.24, (95% CI: 1.05-1.45, P = 0.009) and 1.20 (95% CI: 1.04-1.39, P = 0.013), respectively. Layered meta-analysis showed that in the homozygous comparison model, the Thr307Ala-Asn680Ser polymorphism is a risk factor for male infertility in the white population, with the OR of 1.37 (95% CI: 1.03-1.82, P = 0.003) and 1.21 (95% CI: 1.00-1.47, P = 0.048), respectively.@*CONCLUSIONS@#In the homozygous model (GG vs AA), the FSHRThr307Ala-Asn680Ser gene polymorphism might be a protective factor against male infertility.

OBJECTIVE@#To assess the relation between XRCC3 Thr241Met polymorphism and lung cancer susceptibility of populations in East Asia.@*METHODS@#Related studies of XRCC3 Thr241Met polymorphism and lung cancer susceptibility of populations in East Asia were collected through searching the Pubmed, Embase Library, SPRINGER, CNKI and CSSCI.@*RESULTS@#According to the entry criteria, there were 8 case-control studies in the assessing system and there were 6 321 study cases, including 3 215 patients with lung cancer and 3 106 cases without cancers. Meta analysis results showed the combined OR value of the ratio of genotype Thr/Met+Met/Met and Thr/Thr was 1.03 (95%CI: 0.89-1.20) (P>0.05).@*CONCLUSIONS@#XRCC3 Thr241Met polymorphism may not related to lung cancer susceptibility of populations in East Asia. Allele 241Met did not increase the risk of lung cancer.

Objective To investigate the biodistribution and gamma imaging of 99Tcm-arginine-glutamate-threonine (RET) in nude mice bearing lung cancer xenografts and to explore its feasibility for human lung cancer imaging.Methods RET was labeled directly with 99Tcm.The binding efficiency of 99Tcm-RET with human NSCLC H1299 cells was measured.99Tcm-RET was injected via the tail vein in nude mice bearing H1299 xenografts.The mice(n=32) were sacrificed at different time points:15 min,30 min,1 h,2 h,4 h,8 h,24 h,and 48 h.Organs of interest were excised,weighted and counted by a gamma counter.The organ uptake was calculated as %ID/g.The gamma imaging was performed on 3 mice at 0.5,1,2,4,4.5,5,6 h after intravenous injection of 4.81 MBq 99Tcm-RET.Results The radiolabeling yield of 99Tcm-RET was (93.15±2.02)%,and the binding efficiency of 99Tcm-RET with H1299 cells was (3.56±0.37)%.At 4 h after injection,the uptake of tumor,liver and spleen was (4.96±1.05) %ID/g,(15.89±1.84) %ID/g and (10.83±1.66) %ID/g and the T/NT was 5.70±0.21 for the heart and 12.40±0.11 for the blood.The tumor in nude mice could be best visualized at 4.5-6.0 h.Conclusion 99Tcm-RET might have potential to serve as a lung cancer imaging agent.

ABSTRACT

Se construyó un modelo de simulación dinámica con la intención de explicar el comportamiento del fósforo total en el agua y su relación con el estado trófico del humedal de Jaboque, Bogotá D. C., Colombia. En el modelo se incluyen los flujos de entrada y salida del agua y los aportes de fósforo total en cada una de las tres secciones de la cubeta con características morfométricas y condiciones físicas y químicas del agua muy diferentes. La modelación dinámica se basó en ecuaciones diferenciales y para ella se utilizó el programa computacional Stella 9.1®. En los meses con mayores caudales (época de lluvias), la carga de fósforo disminuye en las tres secciones del humedal, mientras que en los meses con menores caudales (época de sequía), la misma se incrementa. En la sección uno (tercio alto) el valor del índice de estado trófico fue de 45,09 mg/m³ y el tiempo de retención hidráulica de 14 días/año. En la segunda sección (tercio medio) el valor del mencionado índice fue de 35,89 mg/m³ y el THR fue de 34 días/año. Finalmente, en la tercera sección (tercio bajo) los valores fueron 19,5 mg/m³ y 40 días/año respectivamente. Estos valores indican que la primera y segunda sección se encuentran en condiciones de eutrofía y la tercera de mesotrofía. El análisis de sensibilidad permitió estimar que con una carga de 0,62 mg/m³ de fósforo total se podrían mantener las condiciones de mesotrofía en la tercera sección y que para las dos primeras secciones se debería reducir la carga hasta 0,2 mg/m³, para alcanzar un estado de mesotrofía.


A dynamic simulation model to explain the behavior of the total phosphorus in water and its relationship to the trophy status of wetland Jaboque, Bogotá D. C., Colombia, in three sections with particular physical and chemical characteristics is presented. The model includes the inflow and outflow flows of water and total phosphorus contributions in each of the three sections of the wetland with morphometric and water physicochemical conditions very different. The dynamic modelling was based on differential equations; we used the computer program Stella 9.1®. In the months with higher flow (rainy season), the phosphorus load decreases in the three sections of the wetland, while in the months with lower flow (dry season) increases. In section one (third highest) trophy state index was 45.09 mg/m³ and hydraulic retention time of 14 days/year in the second section (middle third) trophy state index was 35.89 mg/m³ and the THR was 34 days/year. In the third section (lower third) values were 5.19 mg/m³ and 40 days/year respectively. These values indicate that the first and second sections are able to eutrophic state and third to mesotrophic. Sensitivity analysis allowed estimating that with a load of 0.62 mg/m³ total phosphorus could be maintained mesotrophic conditions in the third section and for the first two sections should be reduced to 0.2 mg/m³ load to achieve a mesotrophic state.

ABSTRACT

PURPOSE: Simultaneous bilateral total hip replacement (THR) has some advantages, but the efficacy of simultaneous bilateral THR is still controversial in patients with high risk ASA grade. Presently, we compared simultaneous bilateral to staged THR in high-risk patients according to the ASA grade. MATERIALS AND METHODS: We retrospectively compared 60 high risk patients (ASA grade 3 and 4) with simultaneous cementless total hip replacements with a matched group of 60 patients with staged cementless total hip replacements between January, 1991 and June, 2009. Significance was determined to be p-value < 0.05. RESULTS: Postoperative cardio-vascular complication was found in one case of simultaneous THR, and pulmonary thromboembolism was found in two cases of staged THR and in three cases of simultaneous THR. Wound infection was found in three cases of staged THR and in two cases of simultaneous THR. Delirium was found in four cases of staged THR and in five cases of simultaneous THR. Dislocation developed in two cases of staged THR and in three cases of simultaneous THR. Blood loss was higher in the staged group, but the transfusion amount was higher in the simultaneous group. Postoperative ambulation was initiated earlier in staged THR, but at the final follow-up no statistical significance was observed. Admission time and cost were reduced in simultaneous THR. CONCLUSION: It is considered safe to perform simultaneous cementless bilateral THA in patients with high anesthetic risk, if meticulous preoperative evaluation is done.

ABSTRACT

En esta segunda parte del trabajo se focaliza en cómo los estrógenos, con sus diferentes concentraciones a lo largo de las distintas etapas de la vida, por su presencia o ausencia, vulnerabilizan a padecer determinadas patologías neuropsiquiátricas, así como también protegen de algunas otras. En este sentido, se considera que la patología de la mujer debería incluir profundos conocimientos sobre la implicancia que las hormonas sexuales tienen en el desarrollo de determinadas enfermedades neuropsiquiátricas. Se relevan también otros estudios sobre la administración de terapia hormonal de reemplazo, ya que son los procesos neurodegenerativos, como la enfermedad de Alzheimer, los que mayor lugar han tenido entre las investigaciones de las últimas décadas. Allí los resultados se vislumbran promisorios, pues la administración de estrógenos inmediatamente después de la menopausia revela tener efecto en la prevención del desarrollo de estos procesos, lo que no ocurre una vez iniciados los procesos neurodegenerativos. Se incluyen, a su vez, otros trabajos en otras patologías psiquiátricas, en donde se ha evaluado la eficacia de la prescripción de estrógenos, como ser en determinadas formas de depresión mayor (como coadyuvante), con buenos resultados. A pesar de los avances en el campo de la neurociencia y la influencia que ésta ha tenido en el conocimiento de las enfermedades psiquiátricas, es necesario proseguir con las investigaciones en donde se incluyan nuevos fármacos, como ser hormonas sexuales y SERMs, que seguramente traerán aportes promisorios en determinadas patologías neuropsiquiátricas.


The second part of the article focuses on estrogens, with their varying concentrations, throughout the different stages of life, which, whether by their presence or absence, predispose to suffering from certain neuropsychiatric diseases, just as they protect the individual from some others. In this sense, it is considered that female pathology should involve a deep knowledge on the impact that sexual hormones have on the development of certain neuropsychiatric diseases. The article also includes other studies on the administration of hormone replacement therapy, since it is neurodegenerative processes, such as Alzheimer's Disease, which occupied a leading place in investigations during the past decades. In such investigations, outcomes seem to be promising, since the administration of estrogens right after menopause demonstrates to have an impact on preventing the development of these processes, which does not occur once neurodegenerative processes have started. Also included are other investigations conducted on other psychiatric pathologies, in which the efficacy of prescribing estrogens, such as for certain forms of major depression (as adjunctive therapy) are evaluated, with positive results. Despite the advances in neuroscience and its influence on the knowledge of psychiatric diseases, it is necessary to continue preforming investigations that include new pharmacological drugs, such as sexual hormones and SERMs, which will possibly bring about promissory contributions to certain neuropsychiatric diseases.

ABSTRACT

Minimally invasive surgeries, such as laparoscopic, endoscopic and,arthroscopic surgery are currently being used throughout the world. The field of total hip arthroplasty has over 40 years of experience, and minimally invasive total hip arthtoplasty was introduced to reduce the perioperative morbidities. The advocates for minimally invasive total hip arthroplasty will point out the advantages of reduced pain and early ambulation after the operation because of the minimal soft tissue injury. However, we have to review the advantages and disadvantages of minimally invasive total hip arthroplasty to prevent anyharmful effects of this procedure for the patients who under go it.

ABSTRACT

Objective To observe the expressive changes of RhoA and phosphorylated myosin light chain in rat liver tissue undergoing hepatic fibrogenesis.Methods The liver histopathological changes were evaluated by hematoxylin and eosin staining and Masson's trichrome method.Western blot was used to determine the expressions of RhoA and p-MLC(Thr18/Ser19),and reverse transcription-polymerase chain reaction(RT-PCR) to determine the expression of RhoA mRNA.Results With the development of hepatic fibrogenesis,the protein expressions of RhoA and p-MLC(Thr18/Ser19) and the mRNA expression of RhoA were significantly increased.RhoA and p-MLC(Thr18/Ser19)correlated with ?-SMA positively,respectively.Conclusion Rho/ROCK signaling pathways are changed in the process hepatic fibrogenesis.

When adult developmental dysplasia of hip (DDH) patients have developed secondary os- teoarthritis of hip (OAH) with serious clinical symptoms,total hip replacement (THR) is their first choice of treatment.The anatomic structures of the acetabulum and femur in these patients are ahnormal and special,such as small and shallow cup,small femoral head,and narrow medullary cavity at proximal femoral shaft.As a result,the THR for DDH patients is quite different from the conventional ones and is thus highly difficult.We review in this paper the clinical classification of DDH,which is important for its treatment,the difficulties we will have in THR, especially in acetabular reconstruction and femoral prosthesis placement,and also strategies to resolve them.

ABSTRACT

PURPOSE: This paper is proposing an improved CT-free registration method which may provide us an accurate and reliable placement of acetabular cup without CT/MRI images. DESIGN AND METHODS: The proposed method employs a T-bar shaped gauge placed on the anatomical landmarks of the pelvis for the registration. The T-bar shaped gauge has its own LED markers and the position of the gauge is obtained through OPTOTRAK3020 system. These landmark points are the anterior superior iliac spines and the symphysis pubis defining anterior pelvic plane. Two subjects were tested to compare the landmark based registration and the proposed T-bar based registration. RESULTS: The measurement deviations of the pelvic obliquity in the frontal plane, tilt in the sagittal plane, and rotation in the transverse plane were 2.08, 1.41, and 2.51 degrees respectively in the point based registration. The T-bar based registration produced 40% smaller deviations(p<0.05): the pelvic obliquity in the frontal plane, tilt in the sagittal plane, and rotation in the transverse plane were 0.84, 0.81, and 1.17 degrees respectively in the T-bar based registration. There was no outlier exceeding 5 degrees in measurement deviation by the T-bar based registration while the outliers were found by the landmark based registration. CONCLUSIONS: We found that T-bar based CT free registration method is more reliable and accurate than the landmark based registration for the acetabular cup navigation. Also, the new method produced more precise registration(p<0.05). We are expecting some offset error of the new registration method due to the skin thickness existing between the T-bar frame and the bony pelvic frame, which may be compensated if we may accumulate sufficient database of the offset.

ABSTRACT

BACKGROUND: Lipoprotein(a) [Lp(a)] is an atherogenic lipoprotein that is assembled from a low density lipoprotein(LDL) and apolipoprotein(a) [apo(a)]. The variations in Lp(a) concentration tend to be inversely related to the number of kringle IV in apo(a) gene, but other polymorphisms [pentanucleotide(TTTTA) repeats, +93 C/T polymorphism, and Met/Thr polymorphism] of the apo(a) gene are also seems to be related to Lp(a) concentrations. The purpose of this study was to investigate the association of the pentanucleotide repeat polymorphism(PNRP) and Met/Thr polymorphism with the Lp(a) concentrations. METHODS: We studied 197 healthy adults. For genotype analysis of the PNRP and the Met/Thr polymorphism, PCR was performed. Apo(a) phenotyping was performed by SDS-PAGE and immunoblotting. The Lp(a) concentrations were measured by ELISA method. More than two groups were compared using the Kruskal Wallis one-way analysis test. To establish a relationship between gene polymorphisms and Lp(a) concentrations, The linear regression test was performed. RESULTS: Mean Lp(a) concentration was 25.3+/-21.5 mg/dL. Allele frequencies for PNRP, subjects with 8/8 genotype were 114(57.9%) and most frequently observed. The Lp(a) concentrations showed the tendency to decrease as the sum of alleles of PNR increased. Subjects with Met/Thr genotype were 119(60.4%), Met/Met genotype were 71(36.0%) and Thr/Thr genotype were 7(3.6%). CONCLUSIONS: For PNRP, subjects with 8/8 genotype were 114(57.9%) and 8/8 genotype was the most frequently observed. Met/Thr genotype was most frequently observed.
